Abstract: | A promoter is a segment of DNA that controls the expression of a corresponding gene For successful genetic engineering in plants it is necessary to have knowledge of promoters that are used in genetic constructs Methods for optimizing the control of transgene expression have become more important as demand for genetically engineered plants has increased The success of using transgenic technology depends on having an effective promoter that can drive both spatial and temporal gene expression Yet the current number of existing constitutive promoters are limited, and are of viral origin Promoters of plant genes specially from medicinally important species such as Plectranthus amboinicus is essential for developing transgenic plants for the betterment of humankind Constitutive promoters which can robustly elevate transgene expressions in all tissues and at various development phases are especially helpful to express the desired protein in large quantities or develop a specific trait in plants Application of the strong constitutive promoter of viral origin like CaMV35S was found to have certain disadvantages such as its interference in plant development particularly transgene silencing Considering the limitations the promoters that are derived from plants are regarded to be safer and could minimize gene silencing events While there has been a notable rise in the amount of genetic data about plants only a small number of agronomically important plants and model plants have been studied completely However very little information is available about the genome of medicinally important plants such as Plectranthus amboinicus It is an aromatic perennial herbaceous plant that has been widely used in traditional medicines for its curative properties The crop has a great potential because of the expected increase in demand for its pharmaceutical properties that are widely used for managing glaucoma, cardiovascular conditions and cancer types Among the many methods that are available for the isolation of promoters Thermal Asymm |
