Abstract: | Myocardial infarction (MI) is one of the most fatal complications of diabetes mellitus newlinecharacterized by coagulative necrosis of the myocardium, permanent loss of cardiomyocyte newlineand inflammatory cells infiltration in the infarcted region resulting in irreversible loss of newlinecardiac functions and subsequent heart failure. Regeneration of the damaged cardiomyocytes newlineis considered unfeasible with the existing medical options. Accumulating evidence from the newlinepre-clinical and clinical studies demonstrate that stem cell transplantation could be a newlinepotential therapy for treating the injured myocardium and improve cardiac functions in MI newlineheart. Separate stem cell studies in MI and diabetes offer new opportunities in employing newlinecell transplantation in the regeneration of injured myocardium and improving the glycemic newlinecontrol in diabetic infarcted conditions. Among the various available cell types, one cell type newlineof interest is mesenchymal stem cells (MSCs) which are multipotent cells with strong newlineimmunoregulatory potential. The objective of this study was to examine the dose-dependent newlinesafety and efficacy of intravenous administration of human umbilical cord blood derived newlineMSCs (hUCB-MSCs) in chemically induced rats with diabetic infarction. newlineMethods newlineMSCs were extracted and enriched from human umbilical cord blood (hUCB) samples. newlineGrowth rate, enrichment efficiency, and molecular characterization were performed after in newlinevitro enrichment of hUCB-MSCs. Wister rats (weight: 200-250g, males) were newlineintraperitoneally (IP) administered streptozotocin (STZ) injection followed by isoproterenol newline(ISO) to develop diabetes and myocardial infarcted condition. After model development, newlineanimals received intravenous single or double doses of hUCB-MSCs (5 X 106 cells per newlineanimal at each dose) and were followed up to 30 days post-administration.
