Abstract: | Analytical method development paves a way for better efficacy, development and delivery of drug or newlinecombination in this increasing drug market. Analytical Method Development and Validation for newlineSimultaneous Estimation of 5-Flourouracil (5-FU) and Cannabidiol (CBD) 5-FU and CBD had the newlinehighest absorbance at 267 nm and 221 nm respectively. A single wavelength of 235 nm was chosen as newlineper the spectral data of both drugs. The best separation of 5-Florouracil and Cannabidiol was marked in newlinethe mobile phase Water: Methanol in 90:10 ratio and the flow rate of the mobile phase was 1ml/min newlinealong with the total run time of 4 minutes. The retention time of 1.13 minutes for 5-FU and 2.27 newlineminutes for CBD was observed individually and in combination at and#955;max 267, 221, and 235 nm newlinerespectively. Results suggest no change in the retention time of 5-FU and CBD when optimized newlineindividually or in combination. The Correlation Coefficient value (R2) for 5-FU at and#955;max 267nm was newline0.9965, for CBD at and#955;max 221nm was 0.995 and for 5-FU-CBD at and#955;max 235nm was 0.9966 for 5-FU newlineand 0.9998 for CBD indicating the linear relationship. The average retention time for 5-FU was newline1.4284+ 0.026min with RSD of 1.84% and the average retention time for CBD was 2.21+ 0.039min newlinewith RSD of 1.77%. The average area under the curve of 5-Fluorouracil was observed as 1967892.5+ newline31500.34 with RSD of 1.60% and the average curve area of CBD was found to be 3349320.167+ newline35074.94 with RSD of 1.04%. Relative Standard deviation (RSD) values for both retention time and newlinearea under the curve for 5-Fluorouracil and Cannabidiol are below 2%, thus complying with the newlineparameters for system suitability and reproducibility. 5-Fluorouracil and Cannabidiol showed a newlineretention time of 1.44 and 2.2 minutes, respectively, in the 5-Fluorouracil-Cannabidiol mixture and newlineindividually indicating the method is specific. The LOD for 5-Fluorouracil was 2.08 ng/ml at and#955;max newline267nm and for Cannabidiol was 7.6 ng/ml at and#955;max 221nm. |
