dc.description.abstractnewline ABSTRACT newline newlinePharmaceutical co-crystals use crystal engineering principles to create crystalline drug forms that have attracted a lot of interest because they have the potential to solve physicochemical issues with drugs, such as poor aqueous solubility, which can impair drug performance. With desirable qualities like solubility, dissolution, bioavailability, stability, hygroscopicity modulation, pH-dependent solubility properties, photosensitivity, thermolebility, and micromeritic properties, co-crystallization is an emerging technique for designing novel materials. Several pharmaceutical problems that arose during preformulation and formulation development can also be resolved via co-crystal generation. The pharmaceutical industry continues to face a practical problem with weakly basic, poorly soluble active pharmaceutical ingredients (APIs). The most preferable solutions to this problem are crystal forms such as co-crystals, polymorphs, salts, hydrates, and solvates. According to BCS categorization, the majority of APIs are Class II (low solubility and high permeability) chemicals, hence co-crystals have added to the variety of crystal forms at the disposal of pharmaceutical scientists. newline newlineIn this research work omeprazole, lovastatin and nateglinide belong to BCS newlineII class of drugs were selected to prepare co-crystal forms. These co-crystals of APIs newlinewere prepared by using different methods and optimized to produce the best newlinebioavailable and stable forms. In addition to this all physicochemical properties and newlinemicromeritic properties of pure APIs were found to be enhanced by the co-crystal newlinegeneration. newlineOmeprazole is a proton-pump inhibitor used in peptic ulcers, gastro- newlineoesophageal-reflux disorder, Zollinger-Ellison syndrome and in H.pylori infections. newlineOmeprazole is unstable at acidic pH, conditions undergoes degradation in stomach, newlineand insoluble in water, having poor bioavailability. To overcome these major newlinedrawbacks of omeprazole, co-crystallization was attempted, to produce a stable newlinepreparation.
