Title: Nootropic Activity of a Siddha Kayakarpa Drug Poorana Chandrothaya Chenthuram in Experimental Models of Learning and Memory Impairment in Rats

Abstract: Poorana Chandrothaya Chenthuram prepared as per standard Siddha literature is bright red in colour with no specific taste and odour. It complies with the specifications of centuram laid down by the PLIM, Ghaziabad. SEM analysis results revealed that the particle size varies from 1 - 10 m. X-ray diffraction study of the final product showed the peak of gold and mercuric sulphide. The LD50 cut-off value was found to be between 2000-5000mg/kg body weight with reference to the Globally Harmonized System of Classification and Labelling of chemicals. As per OECD test guidelines-423, acute toxicity testing of PCC reveals that the drug can be classified as Category 5 with low acute toxicity hazard. The Indian traditional system of medicine offers a number of safe treatments for central nervous system related disorders such as anxiety and memory loss. These nature-derived treatments are effective and devoid of any untoward effects. The present studies indicate that Poorana Chandrothaya Chenthuram, a Siddha kaya karpa avizhtham, can be regarded as a nootropic agent in view of its facilitator effect on retention of acquired learning. CONCLUSION: Poorana Chandrothaya Chenthuram is a well-processed herbo-mineral formulation containing mercuric sulphide as major component, gold in considerable amount and other micronutrients in minor quantities. Drug doses lower than or equal to 50 mg/kg b.w. in rats did not show any signs of toxicity. The human equivalent dose for high dose given in rat is 480 mg, which indicates that the traditionally recommended dose (65-130 mg for human adult) could be very safe. PCC has been proven in animal models as a worthy nootropic drug with the cholinomimetic action and apoptotic regulator action with significant anti-oxidant property to prevent neuronal cell death. In conclusion, the present study suggests the prospective role of the Siddha formulation Poorana Chandrothaya Chenthuram in providing a holistic relief with high safety margin, from the neurodegenerative disorders of learning and memory impairment
