Abstract: Klebsiella pneumoniae is a common cause of biofilm-based infections, which are intrinsically antibiotic resistant. In this study, the in vitro and in vivo efficacy of DNase I in improving the antibiotic efficacy against K. pneumoniae biofilms was evaluated. Combination with DNase I (16µg/ml) led to four to eight fold improvement in biofilm-eradicating capacity of ciprofloxacin (fluoroquinolone) but not cefotaxime (and#946;-lactam) and amikacin (aminoglycoside). Nearly 99% reduction in biofilm biomass was observed with the combination therapy in a murine model of catheter-associated infection, suggesting the potential utility of DNase I and fluoroquinolone combination in treatment of K. pneumoniae biofilms. newline
