Abstract: In the present research work traditional Siddha formulations Natural and Synthetic Poora parpam were selected and investigated for its physicochemical, toxicological and pharmacological activity against analgesic, anti-inflammatory, antipyretic, antiulcer and hepatoprotective activity in animal models. Preliminary physicochemical analysis such as color, odor, pH, ash value, microscopical and qualitative chemical identification of both Natural and Synthetic Poora parpam proved the genuinity of the drug in accordance with standard ASU guidelines. The angle of repose for Natural and Synthetic Poora parpam seems cohesive and free flowing respectively and hence this property could be the presence of nano particles within the formulation. It was observed that there was a significant increase in the serum levels of AST, ALT and Alkaline phosphatase as a result of CCl4 administration. Treatment with low and high dose of Natural and Synthetic Poora parpam significantly decreased the level of AST, ALT Alkaline phosphatase and Serum Bilirubin in therats belongs to treatment group. Hence it was concluded that Natural and Synthetic Poora parpam have been shown to have hepatoprotective activity. The results of the present work it was concluded that the Siddha formulations Natural and Synthetic Poora parpam have promising analgesic, antiinflammatory, antipyretic, antiulcer and hepatoprotective activity in animals. While comparing the efficacy of the Natural and Synthetic Poora parpam in pharmacological screening it was concluded that the drug Poora parpam prepared from natural source has shown significant higher level of activity when compared to synthetic Poora parpam. Poora parpam is a potent and very safe Siddha formulation, for many diseases like Iduppu Soolai (Lumbar spondilitis), Vatha gunmum (Peptic Ulcer), Suram (Fever), Viranangal (Ulcers), Mega noigal (Venereal diseases), Keel vatham (Osteo arthritis) and Sirangu (Scabies). Clinical studies in human participants can be carried out to establish the safety and efficacy further.
