Abstract: Dhhal3 gene from halotolerant yeast, Debaryomyces hansenii was identified as 1670bp gene translating to 559 amino acids long flavoprotein. 6XHis tagged DhHal3 was bacterially expressed as~73kDa, moderately thermostable protein exhibiting decarboxylase activity. Dhhal3 over expression in D. hansenii exhibited salt stress tolerance phenotype upto 0.4M LiCl and 1.5M NaCl salt concentration while dhhal3 null mutant exhibited a sensitive phenotype. Dhhal3 overexpressing Arabidopsis plant survived 150mM NaCl salt concentration. Overall, Dhhal3 from D. hansenii exhibited the potential to develop salt tolerant crops in the future. newline
