Abstract: Stress is integral part of existence and the mechanism of stress response that involves the activation of Hypothalamo pituitary adrenal HPA axis leading to the release of glucocorticoid has evolved as an efficient adaptive mechanism to meet the energy demands of the moment and to alert the central nervous system for prompt response This adaptive stress response is self limited but if persistent for prolonged periods can lead to disease states Nature has endowed the body with efficient buffer systems to attenuate the stress effects Oxytocin OT and dehydroepiandrosterone DHEA the neuromodulatory hormones are observed to be efficient candidates for stress buffering To assess the effect of prophylactic administration of oxytocin analogue Carbetocin and dehydroepiandrosterone in the attenuation of acute stress in male Wistar rats The study was carried out at CEFTE SRIHER in accordance to CPCSEA guidelines 24 male Wistar rats were segregated into 4 groups with 6 animals in control 6 in stress group and 6 in intervention 1 with prophylactic carbetocin and 6 in intervention 2 group that received prophylactic DHEA before stress 1 hour immobilization was inculcated to animals in stress and intervention groups Blood samples were collected from all animals after 1 hour stress for the estimation of serum corticosterone serum oxytocin and serum DHEA The groups differed significantly in their mean difference in serum corticosterone levels and stress attenuation was significant in both intervention groups while it was with greater significance in DHEA group It could be observed from the findings that prophylactic carbetocin and DHEA administration efficiently attenuated acute stress in male Wistar rats newline
