Development and Evaluation of Novel Colloidal Gel of Nitrendipine for Transdermal Drug Delivery

Abstract

Nitrendipine is a BCS class II drug and belongs to calcium channel blocker class of antihypertensive. The bioavailability of nitrendipine is only 10 to 20% due to extensive first-pass effect. The nitrendipine has side effects like fast heart rate, flushing, chest pain, headache, drowsiness in the patients. All these adverse effects are dose related, so if the dose is reduced the severity of side effects can also be reduced. The reports suggest that co-administration of nitrendipine with the colloidal carriers such as nanoemulsion and niosome enhances the transdermal uptake thus bypasses the hepatic first-pass metabolism and results in enhanced bioavailability which could give reduced side effects. Objectives: The study aimed at development and evaluation of novel colloidal gels like nanoemulsion gel and niosomal gel by using design of experiments for enhancement of bioavailability by transdermal uptake. newline
