Abstract: Neurotoxic disorders account for a significant portion of the diseases that influence the worldwide burden of disease. Parkinson s disease is one of such disease which is linked with environmental toxin exposure. Isocyanates, a highly reactive industrial intermediate used widely in manufacturing plastic product, paints e.t.c. The aim of this study is to delineate the neurotoxic potential of isocyanate in Parkinson s cell model-SHSY-5Y cells. N-succinimidyl N-methylcarbamate; an analogue of methyl isocyanate was used to treat SHSY-5Y cells in dose and time dependant manner. Our findings demonstrate that dose and time dependant N-succinimidyl N-methylcarbamate exposure increases reactive nitrogen species and decreases the glutathione, SOD and catalase level. Further, increased phosphorylated alpha synuclein protein and activation of caspase 3 exert cytotoxicity in SHSY-5Y cells. Our research reveals that widely used N-succinimidyl N-methylcarbamate induce cytotoxicity, apoptosis, oxidative stress as well as protein dysfunction, which might constitute a potential mechanism of the neurodegeneration in Parkinson s disease.
