Abstract: newline ABSTRACT newlineColletotrichum falcatum is one the most known pathogen of sugarcane crop causing red rot newlinedisease. The sugarcane rhizosphere harbors variety of different useful microbes. One hundred newlineeight different mycoflora belonging from the different genera were isolated from the sugarcane newlinerhizospheric soil of resistant varieties (CoPk 05191 and CoLk 94184) and susceptible varieties newline(CoJ 64 and Co 1148) to red rot pathogen. And Trichoderma was found to be predominant in the newlinerhizosphere of sugarcane. Out of 108, fourty one isolates were selected for the study of PGPR newlineand antagonistic related biochemical testsand based on the antagonistic ability against the red rot newlinepathogens (Cf 07, Cf 08, and Cf 09) twelve isolates were selected for further pot and field newlineexperimentation. Among the twelve isolates tested, T16 was found best and recorded higher newlineyield (68.89 t/ha) with highest number of millable canes (76,543) in vivo under glass house newlineconditionas well as under field condition in managing disease. The different level of red rot newlineinfected seed cane viz., 5 %, 10 %, 15 %, 20 %, 25 %, 30%, 35%, and 40 % along with MHAT newlinetreated 10 % infected cane were planted to study the effect of red rot seed borne inoculums on newlinethe performance of variety. MHAT treated 10 % diseased cane seeds showed higher germination newlinepercentage as compared to the untreated seed with 76,456 millable canes per hectare and the newlineyield loss was recorded comparatively less than the untreated cane. Sugarcane juice was newlineoptimized for media preparation and 20 % concentration of sugarcane juice found to be best newlinesuitable in media (SJA) preparation for the growth and sporulation of C. falcatum isolates and newlinealso found equally good as the OMA media to be used as an alternate source of OMA media. newlineCultural characteristics and colony growth rate of thirty five C. falcatum isolates were studied newlineunder light condition and dark condition on OMA and PDA media. Variation among the C. newlinefalcatum isolates was observed in terms of radial growth and col
