Abstract: | newline v newlineABSTRACT newlineCatharanthus roseus (Madagascar periwinkle) is an ornamental plant belonging to the newlineApocynaceae family which is regarded as a rich source of pharmaceutically important newlineTerpenoid Indole Alkaloids (TIAs). Among these, vinblastine and vincristine are of newlineparticular importance because of their wide use in cancer chemotherapy. The present study newlinedeals with the preliminary phytochemical screening which reveals the presence of newlinealkaloids, carbohydrates, flavonoids, phenols, saponins, steroids and tannins in the aqueous newlineand ethanolic extract of the leaf and roots of Catharanthus roseus. The aim of this analysis newlineis to significantly increase the amount of production of secondary metabolites in the leaves newlineand roots of Catharanthus roseus. The plant was exposed to three nanoparticles viz. silver, newlinecobalt and iron in different concentration (0.03, 0.06, 0.10, 0.16, 0.33 g/kg) of soil and newlineresulted in an increase in the amount of secondary metabolites. Qualitative analysis showed newlinethe maximum increase of alkaloid and steroid content in the ethanolic extract of leaves after newlineexposure of 0.33g/kg cobalt nanoparticle (0.257mg/ml) and (44.78mg/ml) respectively. newlineFlavonoid, phenol and saponin was maximum in the ethanolic extract of leaves after newlineexposure of 0.33g/kg (3.406mg/ml), 0.03 g/kg (0.711mg/ml) and 0.33g/kg (92.79 mg/ml) newlineof silver nanoparticle respectively. Carbohydrate and tannin was maximum in the ethanolic newlineextract of leaves after exposure of 0.03g/kg (303.1 mg/ml) and 0.33g/kg (96.36 mg/ml) newlineiron nanoparticle respectively. The HPLC method was done for quantification of newlinevinblastine and catharanthine in the leaves and roots of Catharanthus roseus. The newlineseparation was achieved in 4.5 min for catharanthine and 8.2 min for vinblastine. HPLC newlineanalysis of catharanthine in aqueous and ethanolic extract of leaves showed maximum newlineincrease in 0.33g/kg silver and cobalt nanoparticles from 862.10 µg/ml to 987.00 µg/ml newlineand from 210.22 µg/ml to 670.77 µg/ml in 0.33g/kg silver nanoparticles respectively |
