The purpose of the present research was to investigate the mechanism for improved intercellular and intracellular drug delivery from ethosomes and glycethosomes using visualization techniques and cell line study. In this study we prepare ethosomal gel and glycethosomal dispersion of Berberine chloride and to evaluate for vesicle shape, size, polydispersity index (PDI), zeta potential (ZP), entrapment efficiency (EE), and in vitro permeation studies. Ethosomes were prepared using soyaphosphatidylcholine (1-3%) and ethanol (10-40%) by modified hot method and were characterized and glycethosomes were prepared by soyaphosphotidyl choline, glycerol and non-ionic surfactant. The sizes of ethosomes were found to be in the range of 94.07-297.11 nm and glycethosomes were found in the range 103±13-345±11 nm while PDI ranges from 0.104 to 1.53 in ethosomes and 1.53-0.293 in glycethosomes and ZP was between and#8722;16.6 toand#8722;40.1 mV in ethosomes and-18 to -43mV. Morphology studies showed unilamellar structure under transmission electron microscope and optical microscopy. The EE of ethosomes and glycethosomes was found to be in the range of 42.5% to 90.01%. Ethosomes were further added to carbapol 934P for gel formation, and subsequently, evaluated for their physicochemical properties and glycethosomes evaluated as glycethosomal dispersion. It may be concluded that a Berberine chloride loaded ethosomal gel and glycethosomes dispersion delivery system for herbal medications is a promising technique. newline newline
