Abstract: Thaumatin-like proteins (TLPs) are part of the pathogenesis-related 5 (PR5) family and newlineare involved in stress response. Herein, a total of 109 TLP family genes (93 TLP and 16 newlineTLPK) were identified in the genome of Triticum aestivum. These proteins were grouped into small and long TaTLPs with a conserved thaumatin signature motif. The protein newlineanalyses suggested significant differences between small and long TaTLPs. Evolutionary newlineanalyses suggested the role of duplication events and purifying selection in the expansion of the TLP gene family. Promoter, expression and co-expression analyses revealed the newlinepossible roles of TaTLPs in plant development and abiotic and biotic stress responses, newlinewhich was further validated by qRT-PCR experiment. Recombinant expression of newlineTaTLP2-B in Saccharomyces cerevisiae provided significant tolerance against cold, heat, newlinedrought, and salt stresses. The results depicted the importance of TLPs in cereal crops that would be highly useful in future crop improvement programs. newline newline
