Simulation and validation of efficacy of Cysteamine Hydrochloride in management of Submucous Fibrosis

Abstract

newline Submucous fibrosis (SMF) was recognized as a definitive lesion by Schwartz who described it as a fibrosing newlinecondition in 1952 and due to its predilection for afflicting multiple sites in the oral cavity the disorder was newlinelisted as premalignant/precancerous condition . The proliferation of fibroblasts and newlineoverproduction/accumulation of extracellular matrix (ECM) proteins is well characterized in SMF, however, newlinedespite an extensive knowledge of the cellular and biochemical changes, there remains no effective therapy. newlineOne enzyme that increasingly is associated with ECM is the protein cross-linking enzyme tissue newlinetransglutaminase 2 (TG2). Tissue transglutaminase 2 (TG2) catalyzes the cross-linkage between glutamine newline(Gln) and lysine (Lys) side chains. Cysteamine by virtue of being a transglutaminase 2 substrate, acts as a newlinecompetitive inhibitor of the other amine substrates of this enzyme.
