Abstract: Rhizospheric soil was taken from Sub-Humid and Humid regions of the newlineEastern Rajasthan, India. Soil samples were assembled from Jaipur and Alwar newlinesituated in Sub-Humid region, Kota and Udaipur situated in Humid region of newlineRajasthan state. Samples were processed and used for the isolation of Actinomycetes newlineon Actinomycetes Isolation Agar (AIA) medium. A total of 204 colonies have been newlinetaken from Mother culture plates for the isolation of Actinomycetes from the above newlinementioned four locations. Only 65 isolates were selected for further study, among newlinethem, 9 isolates from Udaipur,16 from Kota, 26 from Alwar, and 14 from Jaipur. newlineMorphological, cultural and biochemical characterization of selected isolates were newlinefollowed by antagonistic activity (primary screening) of selected 15 isolates by disc newlinediffusion and agar well method. Among these 15 isolates, 7 isolates showed newlinepotential to inhibit selected indicator organisms. Fermentation of these seven newlineisolates was performed by keeping them on shaker incubator for two-three weeks newlinefollowed by solvent extraction using four different solvents such as Petroleum Ether, newlineBenzene, chloroform, and ethyl acetate. Crude extracts of different solvents were newlinethen subjected to antimicrobial (AM) activity (secondary screening) against selected newlineindicator organisms on Mueller Hinton Agar (MHA) media. Partial purification of newlinecompounds was executed using Thin Layer Chromatography (TLC) and High- newlinePerformance Thin Layer Chromatography (HPTLC) using different solvent system. newlinePartially purified compounds in the form of bands on TLC plates were scraped and newlinecollected in eppendorf tubes. Active extracts from different solvents and partially newlinepurified compounds by TLC plates were analysed by Gas Chromatography Mass newlineSpectrometry method alongside the detection of active bands having AM activity newlineagainst P. aeruginosa by the bioautography method. GCMS analysis exposed the newlinepresence of 99 major compounds in isolated actinomycetes strains.
