Abstract: newline Citrullus colocynthis has been used since long time for the treatment of various newlinetypes of cancer, hepatic tumour, spleen and eye. Along with that this plant has been used newlineextensively in the treatment of bacterial and fungal infections, ulcers, oxidant stress, newlineinflammatory disorders, hyperplasia and atherosclerosis. Therefore, both plants was newlineselected for present research studies to evaluate in-vitro inhibitory activity on different newlineenzymes like lipoxygenase, hyaluronidase and xanthine oxidase using well established newlinemodels to approve its traditional cases. The botanical identity of collected dried fruits of newlineCitrullus colocynthis and Citrullus lanatus were confirmed before starting the research newlinework by Department of Botanical and Environmental Sciences, Guru Nanak Dev newlineUniversity, Amritsar, Punjab, India with registration number of certificate 1164 and newline1165 dated 11/01/2019, respectively. The various extracts and fractions of fruits of newlineCitrullus colocynthis and Citrullus lanatus obtained from wild areas of different states newlineRajasthan, Punjab and Haryana were prepared using Soxhletion process. The ethyl newlineacetate fraction of Citrullus colocynthis and Citrullus lanatus fruits obtained from wild newlineareas of different states Rajasthan, Punjab and Haryana exhibited strong lipoxygenase newlineinhibition, hyaluronidase inhibitory, xanthine oxidase inhibitory, antioxidant antibacterial newlineand antifungal activities followed by respective methanol extract and acetone extract, as newlinecompared to their respective standard drugs quercetin, tannic acid, allopurinol, ascorbic newlineacid, streptomycin and fluconazole respectively. The ethyl acetate fractions of Citrullus newlinecolocynthis and Citrullus lanatus fruits obtained from wild areas of different states newlineRajasthan, Punjab and Haryana contained larger amounts of total phenols and flavonoid newlineother than their respective acetone and methanol extract. The review of literature newlinesuggested that phenolic and flavonoids compounds exhibited various pharmacological newlineactivities such as antitumor, antiviral, antifungal, antimicrobial, cell fortification, newlinehepatoprotective, antiulcer, antidiabetic and cardioprotective. Therefore, it was concluded newlinethat tested pharmacological activities of present investigations are due to these phenolic
