Abstract: Bud blight disease caused by Groundnut Bud Necrosis Virus (GBNV) is a serious newlineconstraint in tomato cultivating areas. Symptoms associated with bud blight in tomato are newlinechlorotic and necrotic rings on leaves; longitudinal necrotic streaks on stem, petioles and newlinefruit stalks; bronzing of leaves, bud necrosis, chlorotic ring spots on ripened fruits and newlinestunted growth. Owing to its significance, bud blight disease incidence was assessed in newlinefarmers fields in and around Coimbatore, Tamil Nadu. The incidence was noticed up to a newlinemaximum of 84 per cent in the hybrid Shivam 1 and variety PKM 1. The presence of newlineGBNV was confirmed through bioassay and RT-PCR in the symptomatic plants. The virus newlinemechanically transmitted to tomato cv. PKM1 and cowpea cv. Co7 plants showed varying newlinesymptoms of chlorotic and necrotic lesions and systemic infection. newlineReverse transcriptase PCR results confirmed the presence of ~830 bp amplicons newlinepertaining to coat protein gene of GBNV in symptomatic plants collected from the fields newlineas well as in inoculated tomato and cowpea plants. The partial genome sequences of five newlineisolates in BLAST search exhibited 91- 97 per cent identity with groundnut bud necrosis newlinevirus Coimbatore isolate (AY472081), which confirmed the presence of GBNV collected newlinefrom field samples. The partial genome sequences of the five tested isolates were deposited newlinein NCBI database with the accession numbers viz., Thondamuthur (TM1) (MN726915), newlineViraliyur (VL1) (MN726916), Madampatti (MP1) (MN726917), Theethipalyam (TP1) newline(MN726918) and Senanoor (SN1) (MN726919). newlineSimilarly, the partial genome sequence analysis of fungal cultures such as newlineCoprinopsis cinerea, Ganoderma lucidum and Lentinula edodes used in the current study newlineexhibited 99, 98 and 94 per cent similarity with the GenBank isolates of C. cinerea newline(KJ670305), G. lucidum (MF289193) and L. edodes (KF757009), respectively, revealed newlinethrough BLAST search.
