Abstract: | The bisanthraquinones is a class of polyketidic natural product isolated from a large number of newlinefungi and lichens, which possess highly complex molecular structure and display a diverse newlinerange of biological activities. Because of the presence of sensitive and#946;-hydroxyketone groups and newlinemultiple stereo centres in modified bisanthraquinones, it is difficult to synthesize. Biosynthetic newlinestudies have revealed a unified strategy for the biosynthesis of modified bisanthraquinones newlinesuch as ( )-rugulosin, ( )-luteoskyrin, ( )-deoxyluteoskyrin, ( )-rubroskyrin and (+)-rugulosin newlinein nature. This inspired us to develop a biomimetic, chemoenzymatic method for the synthesis newlineof bisanthraquinones, which is described in thesis. newlineIt involves reductive dearomatization of anthrol precursor with NADPH dependent anthrol newlinereductases such as MdpC, 17and#946;-HSDcl or anthrol reductase of Talaromyces islandicus (ARti) newlineto obtain a putative biosynthetic intermediate, which is being dimerized and modified to obtain newlinevarious bisanthraquinones in just 3-4 steps starting from anthraquinones. newline |
