Abstract: Chronic diseases cause remarkable impairment in patients and signify a substantial economic burden on healthcare resources. Dysbiosis is the causative factor for the various chronic diseases such as liver inflammations, obesity, Type-2 diabetes mellitus, and other diseases such as colon cancer, obesity induced chronic kidney disease (CKD), compromised lung immunity, and some of CNS disorders. Normobiosis is the precise approach to the prevention of chronic diseases by using prebiotics and probiotics in the diet. newlineThe use of natural remedies to alleviate chronic disease has a long history. Existing synthetic agents for the treatment of chronic disease are inadequate and known to have many undesirable side effects, therefore several attempts are being made to discover reliable preventive agents from natural origin. newlineIn the present study, a probiotic was isolated from sauerkraut and identified as a Pediococcus Pentosaceus DSM20336(T) by gene sequencing and studied its phenotypic and genotypic characteristics, on the basis of these characteristics it was concluded that Pediococcus pentosaceus is an ideal candidate as probiotics Which is used for further study. Prebiotics are a group of nutrients that are degraded by gut microbiota considering the health benefits. Cucurbita maxima and allium sativum extract as prebiotics were selected and screened for their prebiotic and growth stimulatory effects in vitro. newlinePolyherbal formulation of sauerkraut juice and Hydroalcoholic extract of Cucurbita maxima and allium sativum was designed by using design expert software and optimized formula in higher concentration and lower concentrations were used for to evaluate the antidiabetic and antiobesity potential in High-fat diet and Streptozotocin (STZ) induced diabetes and obesity. newlineThus, the present study showed that the Polyherbal formulation in higher concentrations has remarkable antihyperlipidemic, antiobesity and hypoglycemic activity in high-fat diet and STZ-induced diabetic rats. newlineThis hypoglycemic and antiobesity effect may be d
