Abstract: Traditional Poly Herbal Formulation (PHF) with six medicinal plants Ximenia americana, Cassia auriculata, Emblica officinalis, Terminalia belerica, Smilax china and Strychnos potatorum as ingredients was selected for the study. The aim of this study is to develop a traditional anti-diabetic polyherbal formulation and evaluation of Pharmacognostical, Phytochemical and Pharmacological aspects of anti-diabetic activity for developing a standard monograph and it is also aimed to develop and evaluate a formulation of oral dosage form tablet from traditional kasayam to emphasize patient compliance. Developing and evaluating a convenient oral dosage form, tablet improves the shelf life and patient compliance. Four different batches were formulated. Out of the four batches, Batch No. PHF/004/19 has showed excellent results in all the parameters and in stability studies also. Modernization of traditional formulation can be achieved from this research work which will benefit in the treatment of people with diabetic mellitus. The results of pharmacognostical and phytochemical standardization were helpful in establishing quality control standards and purity of drugs. So, the present formulation was selected for film coating, quantification of gallic acid and for stability studies.Accelerated stability studies did not show any significant change in the physical properties, total phenol content, disintegration and dissolution for the period of six months. So, novel antidiabetic formulation ensuring quality, safety and efficacy was developed using traditional medicine. The findings from the present study are the first experimental data providing the scientific evidence to rationalize and support the traditional use of this polyherbal formulation for the treatment of diabetes mellitus. The well-developed Tablet dosage form of PHF ensures increased shelf life of the formulation as well as patient compliance. Hence, this polyherbal formulation has the potential to be accepted as an antidiabetic formulation globally.
