Abstract: | Extract of Curcuma longa, Piper nigrum and Berberis aristata is reported to be useful in stomach newlineailments and has been shown to possess antiulcer activity. Since, treatment for peptic ulceration newlineincludes a consumption of multiple herbs displays synergistic impacts compare to single herb newlineagainst peptic ulcer. This project work was defined to retain the herbal extracts based newlinegastroretentive formulation in stomach for better antiulcer activity. Hence, formulation and newlineevaluation floating tablet of spray dried extract of Curcuma longa, Piper nigrum and Berberis newlinearistata was carried out. Prior to development of gastroretentive formulation, pharmacological newlineactivity and simultaneous HPLC method were performed. From the pharmacological activity, newlinecombined methanolic extracts of Curcuma longa (250mg/kg), Piper nigrum (125mg/kg) and newlineBerberis aristata (125mg/kg) significantly diminished the ulcer index compared with Ranitidine newlinein ethanol and pylorus model. HPLC method was carried out on Spectra system, P-400 HPLC newlineusing CurosilPFP 5and#956; as stationary phase and mobile phase containing Methanol: 10mM newlineAmmonium acetate with acetic acid (pH 3) (75:25%v/v) at flow rate of 1ml/min using UV newlinedetection at 254nm. Hence, developed method was accurate, precise, simple, sensitive, selective, newlinerobust and rapid. The floating tablets were formulated using hydrophilic polymers as Hydroxy newlinepropyl methyl cellulose (HPMC K4M and HPMC E15), Sodium CMC and Sodium bicarbonate newlineas a gas generating agent. The formulated tablets were evaluated for the quality control tests such newlineas weight variation, hardness, friability, swelling index, floating lag time, total floating time and newlinein vitro drug release study. The results of the present study clearly indicate the HPMC K4M and newlineHPMC E15 (Batch F-11) in appropriate concentration shown buoyancy of tablet for 8 hours along newlinewith the sustained drug release in 0.1N HCl as dissolution media. newline |
