Abstract: Filamentous fungi from the genus Aspergillus are of high importance for the production of organic acids. Itaconic Acid (IA) is considered as an important organic acid for the production of synthetic fibers, resin, plastics, rubber, paints, coatings, adhesives, thickeners and binders. Currently, microbial production of itaconic acid using diverse microorganisms from alternative substrates received a greater potential. Biofuel production has become an important alternative resource towards the replacement of ancient fossil fuels. Algal sources serve as a best source for the generation of fatty acids which can be converted into biodiesel through process called trans-esterification. Subsequently, residue algal biomass and crude glycerol are accumulated in enormous quantity. Biosynthesis of itaconic acid using isolated microorganism from algal biomass hydrolysate and purified glycerol was the most important intention of this thesis. newlineOriginally, isolation of fungal species was performed using Potato Dextrose Agar (PDA) medium. The isolated fungal strains were subjected to screen for the production of itaconic acid by using formation of natural acids method and quantification of itaconic acid. The fungal strain which showed maximum itaconic acid was subjected to identification of respective microorganism through 18 S rRNA sequencing methodology. cis Aconitic acid decarboxylase (CAD) enzyme activity was measured during the growth of the microorganism. Among the isolated fungal species, RGB 14 strain showed maximum production towards itaconic acid during the initial screening steps and the respective fungal species was identified as Aspergillus niveus. newline
