dc.description.abstractAim: newlineThe aim of this study was to evaluate antimicrobial and cytotoxicity properties newlineof nanosilver varnish, chitosan Nano particle and combination of both nano newlinesilver-chitosan nanocomposite varnish against Streptococcus mutans, newlineStreptococcus aureus, Lactobacillus sp and Candida albicans compared with a commercially available dental varnish by using them in vitro and in vivo newlineanimal models. newlineMaterials and methods: newlineFor in vitro study the organisms used were Streptococcus mutans, newlineStaphylococcus aureus, Lactobacillus sp and Candida albicans. Agar well newlinediffusion method was used to assess the antimicrobial efficacy of the newlinenanoparticles and the nanoparticle incorporated dental varnish at 25 µL, 50 newlineµL, 100 µLand 200 µL. To assess the cytotoxic effect, brine shrimp lethality newlineassay was used. For animal study, 30 Wistar albino rats were treated, and newlinesubmitted to a cariogenic challenge. After 4 weeks, total microbial count, newlineStreptococcus mutans, Staphylococcus aureus and Lactobacillus sp oral newlinepresence were evaluated. newlineResults: newlineZone of inhibition was found to be highest at 200 µL concentration of all the newlinegroups against Streptococcus mutans, Staphylococcus aureus, Lactobacillus newlinesp and Candida albicans. The cytotoxic activity at 5 µL and 10 µL was 0%. newlineThe maximum cytotoxicity was seen at 80 µL where 30% of the nauplii died. newlineFor animal study, the nanocomposite showed a statistically significant newlinereduction in the percentage of total microbial count, Streptococcus mutans, newlineStaphylococcus aureus and Lactobacillus sp compared to the positive control newline(p lt 0.05). newlineConclusion: newlineFindings from this study suggest that silver nanoparticle, chitosan newlinenanoparticle and nanocomposite extracts has the potential as an newlineantimicrobial agent and has less cytotoxic effect on brine shrimp. newlineNanocomposite showed reduction in the percentage of total microbial count, newlineStreptococcus mutans, Staphylococcus aureus and Lactobacillus sp in vivo. newlineKeywords: Antimicrobial, Cytotoxicity, Dental Caries, Dental varnish, Mutans newlinestreptococci, Nanocomposite
