Abstract: Abstract newlineBackground: newlineThe success of endodontic treatment broadly depends on the complete newlinedebridement of microbial flora from the root canal system. This can be achieved newlineby thorough disinfection of the canals which employs the usage of intracanal newlinemedicaments to a great extent. Intracanal medicaments aid in the inhibition of newlinemicrobial growth to a large extent. An effective intracanal medicament can newlinereduce the microbial load in the root canal. newlineAim: newlineThe purpose of this study was to evaluate the effect of two intracanal newlinemedicaments against microorganisms in the infected root canals. newlineMaterials and methods: newlineIn a prospective randomized clinical trial, 40 patients were selected with mean newlineage between 18 to 70 years according to inclusion and exclusion criteria and newlineintracanal samples were collected. Based on block randomisation patients were newlinedivided into Group A: Diclofenac sodium and Group B: Calcium hydroxide. After newlineroutine root newlinecanal preparation, in Group A and Group B cleaning and shaping was done newlinefollowing the standard protocol until 6% 25 Protaper Gold rotary files. A second newlinesample from the cleaned and shaped root canal was collected. Intracanal newlinemedicament was placed based on the assigned group using lentulo spirals and newlinethe patient was recalled after seven days. A third sample from the root canals newlinewas obtained and obturation was completed. The microbial load in the root newlinecanal samples before and after the treatment was evaluated by RT-PCR/qPCR newlineand analysed statistically. newlineResults: newlineBoth the groups showed a decrease in the total microbial count before the newlinetreatment, after cleaning and shaping was performed and after the medicament newlinewas placed in the canals for 7 days. There was not much of a significant newlinedifference in values before the start of the treatment and after the completion of newlinethe treatment in both the groups. newlineConclusion: newlineWithin the limitations of the study, Diclofenac sodium can be used as an newlineintracanal medicament.
