dc.description.abstractThe present work is performed with a thorough and prior literature search,which has led to understanding that Phytate is known for its anti-neoplastic activityinvariouscancers,buttheunderlyingmechanismsarelessunderstood.WethereforeexaminedpureInositolHexaphosphate(IP6,phytate)asadietaryingredientprovidedpost-initiation,foritsabilitytoinhibitdysplasticaberrantcryptfoci(ACF)formation, and their progression to colon adenoma/adenocarcinomas viz. a viz. therole of altered iron homeostasis, in F344 rats using azoxymethane (AOM) as acarcinogen. Treatment with 1 or 2% IP6 was initiated after 20 weeks of AOMadministration and continued for 16 weeks. IP6 suppressed ACF formation andinhibitedtheirprogressiontotumorsinadose-dependentmanner.Phytatesignificantly ameliorated the tumor and inflammatory markers such as proliferatedcellnuclearantigen(PCNA),and#61538;-catenin,cyclooxygenase-2(COX-2),whoseexpression was up regulated with azoxymethane. Systemic iron status indicatorsincluding hemoglobin, serum iron, ferritin, and hepcidin levels suggested a cleardepletion of body iron stores with phytate supplementation. Apoptotic indices, suchasTUNELstainingandcaspase-3activitywerepromotedbyIP6inAOM-administeredcolonepithelium.Thus,dietaryphytateconsumptioneffectivelyreduced the tumor incidence and multiplicity even in a post-initiation model ofazoxymethane, with a decreasein bodyiron stores. newline
