Abstract: Metabolic syndrome is a lifestyle disease, where diet and gut microbiota play a prodigious role in its initiation and progression. Prophylactic bio-interventions employing probiotics offer an alternate approach towards attenuating its progression. In the present study, anti-inflammatory lactic acid bacteria were isolated, screened, identified and assessed for prebiotic metabolizing potential. Further, supplementation of synbiotic (selected probiotic isolate Lactobacillus pentosus GSSK2 and prebiotic isomalto-oligosaccharides) for 12 weeks to animals fed with high-fat diet ameliorated anthropometric parameters, glucose clearance, lipid biomarkers, alleviated oxidative stress, restored the gut microbiome, histomorphology and modulated inflammatory, lipid and glucose metabolism genes in adipose tissue, colon and liver. Thus, the study highlights the protective potential of synbiotic in ameliorating diet induced metabolic syndrome.
