Abstract: The main objective of the present study is to Estimate risk factors and newlinemicrobiological profile of blood samples of septicaemic patients in neonatal ICU of newlinea tertiary care hospital Dehradun in Uttarakhand. The aim of the study is as to find newlineout the antibiogram of the selected isolates, frequency of AmpC gene producing beta newlinelactamases, assess various phenotypic techniques for ESBLs detection and newlinecharacterise the genotype of the isolates. newlineThis Cross Sectional study was carried out in the Department of newlineMicrobiology of College and Hospital, Dehradun in Uttarakhand. Three eighty two newlinesamples were collected assuming 58% ESBL producers among the isolates. The data newlineregarding age, sex, risk factors, patient identification number were collected from newlinethe Medical Record Department. Blood samples were collected from the patients by newlinevenipuncture under aseptic conditions. The data were entered and analyzed using newlineStatistical Package for Social Sciences (SPSS, version. newlineIn the current study combined disc diffusion test (CDDT) recommended by newlineCLSI was considered as the standard method. The validity of all other tests was newlinecompared with the standard tests. newlineIn the present study double disc approximation test detected 17 isolates as newlinepositive for ESBL producers which were 100% truly positive by CDDT. Twelve newline(12) isolates were detected as negative, out of which only 05 (41.67%) were true newlinenegative by CDDT. As 17 (70.83%) out of 24 positive isolates were detected newlineconfirmed by double disc approximation test. By using Disc antagonism test for newlineinducible AmpC ß-lactamases and Ezy MIC strip test (EM079 and EM081) newline(Himedia, Mumbai), n=2 (6.89%) isolates were detected for co-production of both newlineESBL and AmpC beta-lactamase among the 29 isolates. One (n=1/16) (6.25%) of E. newlinecoli isolated from LONS case and one (n=1/13) (7.69%) of Klebsiella spp isolated newlinexvi newlinefrom EONS case produced ESBL as well as AmpC and#946;-lactamases. By using both newlinethese methods, 01 (6.25%) E. coli and 01 (7.69%) Klebsiella spp isolates were newlineid
