dc.description.abstract | The aim of present investigation was to develop and evaluate in situ nanogel based topical spray for the treatment of psoriasis. Polyvinyl alcohol (PVA) and hyaluronic acid (HA) with immunosuppressant drug in situ nanogel can apply on inflamed skin by topical delivery. Fourier transform infrared spectroscopy (FTIR) and Differential scanning calorimetry had employed to study drug-excipients incompatibility. Analytical method was performed using UV spectrophotometer. HA-PVA nanogel was successfully prepared using newlineimmunosuppressant drug, polyvinyl alcohol(PVA), Hyaluronic acid(HA), Glutaraldehyde (chemical cross linker) by chemical crosslinking method and evaluated for viscosity, pH, particle size, zeta potential, percent drug entrapment, gelling time, skin retention and permeation study by HPLC method, in-vitro drug release, in vivo study and stability study for nanogel formulation. Lyophilization process performed for nanogel convert into powder dosage form. Flow property, Drug content uniformity, FT IR and DSC study was peromed as evaluation parameter of lyophilized nanogel formulation. For optimization, 32 full factorial designs for Dimethylfumarate containing in situ nanogel have been used to study the effect of newlineindependent variables on dependent variables. For optimization, 32 full factorial designs for cyclosporine A containing in situ nanogel have been used to study the effect of independent variables on dependent variables. All evualation parameter was found satisfactory of both formulation. Stability study shows developed lyophilized Nanogel was stable at 25°C± 2°C/60%±5%(refrigerator condtion) at accelerated condition after 6 month. The present study demonstrated that in situ nanogel based topical spray has good effect on psoriasis skin pH than other dosage form. Topical Spray cover enlarge portion of the skin without irritation and given effect on the skin. | |
