Abstract: | The objective of this study was to prepare and evaluate an enteric coated capsule containing Capecitabine and Oxaliplatin mucoadhesive microspheres for the treatment of colon cancer. Antineoplastic drugs such as Capecitabine and Oxaliplatin can be administered orally. HPLC newlinehas created an analytical approach for the estimation of pharmaceuticals. The drug: polymer ratio (1:2), inlet temperature 170°C, and crosslinking agent 0.5 ml 1 % Gluteraldehyde solution were all optimised using the Box Behnken design. The microspheres were successfully prepared by newlineusing the spray drying technique. The results of optimised Capecitabine microspheres were newlineobtained as Particle size 87.91µm ± 0.274, % yield 57.21± 1.5, % Mucoadhesion 57.21± 1.5, % entrapment efficiency 82.16± 0.725 and swelling index 2.8 ± 0.19. The results of optimised Oxaliplatin microspheres were obtained as Particle size 99.88µm±0.034, % yield 56.0± 0.088, % Mucoadhesion 87.0± 0.80, % entrapment efficiency 82.61±0.085 and swelling index 2.71±1.17.Drug content of Capecitabine and Oxaliplatin in filled capsule was found to be 94.67% ±0.32 and 93.45%±0.712 respectively. % Drug release of Capecitabine and Oxaliplatin in PBS pH 7.4 was found to be 94.83±0.22 and 96.94±0.11 respectively after 8 hrs. The MTT assay was used to evaluate the % viability of Capecitabine and Oxaliplatin mucoadhesive newlinemicrospheres containing capsule formulations on the Caco-2 cell line. When compared to API, the formulation resulted in a significant rate of cell death within 48 hours and lower cell viability. The enteric coated capsule remained stable at 0±2°C/75±5% in accelerated conditions after 6 months, according to the stability analysis. The current investigation found that enteric newlinecoated capsules containing produced mucoadhesive microspheres have an anticancer effect on colon cancer cells. newline newline |
