dc.description.abstractThe aim of the study was to perform phytochemical and pharmacological evaluation of the newlinebioactive fractionates obtained from Sida cordifolia Linn. and Ventilago calyculata Tul. for newlinewound healing potential against aspirin and dexamethasone retarded wound healing process newlineand to understand the probable unknown cellular and molecular mechanism of their action. newlineWe evaluated the wound healing property of successive extracts (petroleum ether, ethyl newlineacetate, methanol and aqueous) of Sida cordifolia and Ventilago calyculata against newlineDecdan®/Disprin® (dexamethasone/aspirin)-induced retardation of wound healing in rats. The newlinevarious extracts of selected plants were characterised by determining the various newlinephytochemicals and quantifying the total phenolic content and flavonoidal content by highthroughput newlineassays. newlineIn order to know the probable mechanism of action of the successive fractionates the newlineantioxidant activity was assessed both by in vitro (DPPH-assay) and in vivo methods in newlineSaccharomyces cerevisiae-wild type (BY4743) and knock-out strain (and#916;trx2). The cell newlinemembrane integrity/amount of mitochondrial ROS was determined by treating the strains newlinewith extract/standard in presence of H2O2 and Propidium iodide (PI)/MitoSOXTM Red. newlineThe statistical study revealed that in excision and incision wound models all the formulations newlinehad significant (plt0.01) wound healing potential compared to the negative control. However, newlineaqueous extract formulation of S. cordifolia (HF4) and V. calyculata (HF8) were found to be newlinesuperior to all other treatment groups against Decdan®/Disprin® respectively. Several newlinepharmacologically important phytoconstituents were also detected in the extracts which newlinemight be responsible for potent wound healing activity. Regarding its mechanism of action, newlineour findings report that aqueous extract formulation of S. cordifolia (SCA) was able to rescue newlinethe and#916;trx2 strains from stress, it can be inferred that it might be able to induce the enzyme thioredoxin-II to restore redox homeostasis.
