Abstract: | Treatment methods for cancer that are widely being utilized affect both normal and cancerous cells. Normal tissue toxicity with better tumour control can be attained using superparamagnetic nanoparticle as radiosensitizer. PEG-coated Fe3O4 NPs were synthesized by chemical co-precipitation method and characterized by studying their size, structure, functional group, stability, magnetization, and cytotoxicity using different methods. The potential role of SPIONs as radiosensitizer on HT 29 cell lines for different concentrations and radiation doses of 6MV photon beam is assessed. XRD, FTIR and TGA results shows that Fe3O4 NPs have been coated with PEG during the synthesis. Zeta potential study shows that NPs are stable. coated Fe3O4 has a greater hydrodynamic size than bare Fe3O4. TEM micrographs exhibited that NPs are roughly spherical with size in range of 10-20 nm. Saturation magnetization value of coated and bare Fe3O4 also confirms coating and shows superparamagnetic behavior. Cytotoxicity study indicated that coated Fe3O4 is biocompatible on L929 and toxic on MCF-7. Combination of radiation and PEG coated SPIONs results in increasing cell killing of HT 29 as compared to radiation alone. The goal of radiotherapy i.e. achieving better tumor control while minimizing damage to healthy tissue can be fruitfully achieved using PEG coated SPIONs. newline |
