dc.description.abstractAlkaline pectinase is the utmost significant industrial enzyme of the bio scouring process. By newlineconsidering bio scouring of cotton, 30 microbial isolates from fruit and vegetable waste-rich dump newlinesoil of Solang Valley and Vasishta (Manali, Himachal Pradesh, India) were isolated and screened for newlinethe alkaline pectinase production in the current research work. Only four isolates P3, P16, P21, and newlineP27 were capable to produce extracellular alkaline pectinase at pH 9. Further by applying submerged newlinefermentation, the alkaline pectinase production was quantitatively screened. The most efficient isolate newlinewas P3 identified as Bacillus tropicus, based on morphological, biochemical, and molecular newlinecharacterization. Molecular characteristics confirmed by 16S rDNA sequence analysis. The newlinenucleotide sequence of the isolate was novel with a 97% similarity index and submitted to the newlineGenBank with accession number MK332379. This isolate was characterized as Bacillus tropicus newlineMCCC1A01406 based on morphological, biochemical, and molecular analysis. It was found to be newlineGram-positive, rod-shaped (2.0-2.5 pm), spore-forming, aerobic, non-motile, catalase, and oxidasepositive newline newlinebacterium. Bacillus tropicus MCCC1A01406 also produced other hydrolytic enzymes such newlineas lipase, amylase, xylanase cellulase, and protease but lacked chitinase and tannase. Pectinase newlineproduced by Bacillus tropicus MCCC1A01406 was extracellular. This strain grew well on most of newlinethe carbon sources used but produced acid only in glucose, fructose, sucrose, galactose, and maltose. newlineThe isolate grew well between pH and temperature range of 5.0-11.0 and 25-45°C respectively. This newlineisolate was capable of hydrolyzing tributyine, xylan, starch, casein, and carboxymethyl cellulose, but newlinedid not hydrolyze tannic acid, chitin, and urea.Of the various complex media tested, yeast extract newline(1%, w/v) supplemented with pectin (0.25%, w/v), was found to support maximum pectinase newlineproduction.
