Abstract: Objectives-To quantify the parameter of Nischandratva of Abhrak Bhasma, explore cell mediated immune response, anti-proliferative, chemopreventive effect on breast cancer and heavy metal analysis of Rasasindoor and Shataputi Abhrak Bhasma. newlineMethod-Rasasindoor and Shataputi Abhrak Bhasma were prepared at D.Y.Patil School of Ayurveda pharmacy. Spectroscopic analysis of Nischandratva of Abhrak Bhasma of various Putas was done with UV-Visible-Infrared spectroscopy. Rasasindoor and Shataputi Abhrak Bhasma were evaluated for cell mediated immune response by performing the Delayed type hypersensitivity animal study. The test drugs were subjected to invitro anti-cancer activity on human breast cancer cell lines MCF-7, in varying concentrations using MTT assay. Chemoprevention effect of the test drugs was analysed with DMBA-induced breast cancer animal model. Heavy metal analysis in chemopreventive study was done by subjecting blood and liver tissue to ICP-MS and ICP-AES. newlineResults-Spectroscopic analysis of Nischandratva of Abhrak Bhasma established a non-linear inverse relationship between incinerations and percent reflectance in all the three zones of sunlight. Results of the in-vitro immunomodulatory study indicate that the test drugs potentially supress cellular immune responses by inhibiting SRBC induced Delayed type hypersensitivity in mice. The test drugs showed anti-proliferative activity on human breast cancer cell line MCF-7 in concentration dependent manner. The test drugs significantly prevented development of breast tumors in DMBA-induced breast cancer. The results of heavy metal analysis study have shown that the levels of heavy metals in blood and liver tissue are all within the normal permissible limits. newlineConclusion-Number of incinerations have an inverse relation with UV-Visible-Infrared light. Test drugs have a suppressive effect on cell-mediated immunity, have a significant anti-proliferative and chemopreventive efficacy on breast cancer and do not lead to heavy metal accumulation in blood or liver tissue
