Abstract: | Type 2 diabetes is a difficult and common condition in India. The interaction of environmental and genetic factors disrupts several physiological processes, creating T2DM. Its participation in the disease has been extensively studied in the quest for an early predictor to avert financial hardship and subsequent problems. Despite extensive study, identifying and correlating genetic variables with T2DM remains difficult because to ethnic differences. Before using the candidate gene approach to investigate the association of CAPN10 (rs3842570), FABP2 (rs1799883), GLUT2 (rs5393), TCF7L2 (rs7903146), and ENPP1 (rs1044498) with T2DM in the considered population sample, the study assessed the validity of anthropometric and biochemical parameters as predictive markers in Navi-Mumbai. Genetic selection was based on gene involvement in glucose metabolism, insulin secretion/action and/or lipid metabolism. newlineThe proposed in vitro techniques were tested in silico for efficacy. 300 people, both diabetic and non-diabetic, were chosen at random. Anthropometric and biochemical measurements were compared between control and cases to find a significant correlation. Hip circumference was observed to be increased in diabetics, regardless of age, BMI, or waist size. The diabetic group had significantly higher mean fasting blood glucose, triglyceride, TG:HDL ratio, and VLDL levels than the healthy group, but cholesterol, HDL, and LDL levels were non-significant. Females had a slightly higher frequency and T2DM risk increased with age. The PCR-RFLP method, also known as Cleaved Amplified Polymorphic Sequences (CAPS), was used to genotype the samples. The Hardy Weinberg equilibrium was confirmed for all five genes. TCF7L2 (rs7903146) was shown to be related with T2DM, with the C allele being protective and the T variant being associated with increased risk. The remaining four genes had no significant relationships. New GLUT2 (rs5393) polymorphism primers for PCR-RFLP genotyping were created. newline newline |
