dc.description.abstract | ABSTRACT newline newlineThe present study investigated the effect of 5 methoxyflavone on central nervous system functions in wistar rats by employing three behavioural models such as open field test, potentiation of pentobarbitone and ether anaesthesia. Mice were employed to identify the mechanism of hypnotic effect of 5 methoxyflavone by administering suitable interacting chemicals.Docking studies were also carried out to identify the molecular interactions as well as the binding affinity of 5 methoxyflavone.The potential general anaesthetic effect of 5 methoxyflavone upon i.v administration was carried out in three animal species (viz) mice, rats and rabbits.Maximal electroshock and pentylenetetrazole induced seizures in mice were employed to ascertain the anticonvulsant effect of 5 methoxyflavone. The anxiolytic like effect of 5 methoxyflavone was tested on two unconditioned models of anxiety namely light dark box and elevated plus maze test in mice.A significant and dose-dependent reduction in locomotor activity was evident in rats and a significant increase in the duration of sleep was observed after 5-methoxyflavone treatment. It also revealed the participation of GABAA, adenosine, glycine and NMDA receptors in the hypnotic effect of 5 methoxyflavoneby in-vivo and in-silico studies. The study also revealed an ultra-short acting general anaesthetic effect of 5 methoxyflavone in all the three animal species tested.5 methoxyflavonetreatment significantly attenuated the seizures induced by maximal electroshock and pentylenetetrazole. A significant increase in the time spent in the light compartment in the light dark box test and also an increase in the percentage of entry and the time spent in open arms in an elevated plus maze was observed. The results of the present study revealed a sedative-hypnotic, general anaesthetic, anticonvulsant and anxiolytic like effect of 5 methoxyflavone in various experimental animal models. newline newline | |
