Biosynthesis Of Gold Nanocomposites And Their Studies On Anticancer Applicaton

Abstract

Gold nanoparticles biosynthesis has been achieved by Glycyrrhiza glabra, Catharanthus roseus and Scoparia dulcis, by cost-effective and rapid process. Based on the characterization results, Scoparia dulcis was found to be more stable and were selected for further studies. The GC- MS analysis of S. dulcis synthesized AuNPs reveals the phytoconstituents present in it. The results of MTT assay shows that the AuNPs exhibits considerable anticancer activity against HepG2 cells with the EC50 value in 82µg/mL. The results of FITC-labelled Annexin V/ PI assay show a dose-dependent increase of cell death in AuNPs treated HepG2 cells. The treatment of AuNPs up-regulated the Caspase- newline3 in HepG2 cells. In H2DCFDA staining assay, AuNPs were found to induce toxicity in zebrafish embryos with LC50 value of 12.501%. Moreover, AuNPs ameliorated/restricted the progression of neoplastic changes in the DENA induced hepatic cancer in zebrafish. The in-vitro newlineand in-vivo studies suggest that S.dulcis synthesized AuNPs exhibits its newline newline newlinepotential to be an anticancer agent for the Hepatic cancer. newline
