Abstract: | One of the leading causes of death of most of the people, reported so far, globally is cancer and there is a very high uncertainty that this will be improved, as the number of cases keeps on increasing gradually. Several kinds of medicines are available in the market to treat the different types of cancer but these medicines safety and efficacy profiles remain questionable. One of the main problems that appear in cancer chemotherapy is the toxicity of the established drugs. However, naturally, derived products have been proven to be safe and effective in the treatment and management of cancers. Hence, in the current trend, most of the researchers are doing their research on bio-based natural products to treat cancer. Many naturally derived products or their analogs have been identified as potent anti-cancer agents and similarly, the anti-cancer potential of various bio-compounds is being identified day by day. Considering this data on the increasing importance of bio-compounds, we have also attempted to study the efficacy of Hispolon, Limonene, and a combination of Doxorubicin in the treatment of colon cancer. newlineIn this current study, we investigated the combinatorial anti-cancer effect of Doxorubicin, Hispolon, and D-Limonene against colorectal cancer cell lines, which was determined by MTT assay. The antioxidant enzyme activity of various combinations of the tri-drug on COLO-205 and HCT-116 cells was performed. The activity of antioxidant enzymes, SOD, Catalase, GPx, and GST were measured. The free radical scavenging activity of Doxorubicin, Hispolon, and D-Limonene performed on DPPH, ABTS and nitric oxide free radicals were measured. The RTPCR and western blotting methods determined the gene and protein expression levels of selected genes and proteins upon introduction of Doxorubicin, Hispolon, and D-Limonene to the COLO 205 and HCT 116 cells. All of the tests were carried out three times in triplicates. |
