dc.description.abstractAlcoholic Liver Disease (ALD) has a complex process that includes a wide spectrum newlineof hepatic lesions from steatosis to cirrhosis. Cell injury, inflammation and oxidative stress are the key drivers of alcohol-induced liver injury. Alcohol abuse causes accumulation of newlinereactive oxygen species, which in turn causes lipid peroxidation of cellular membranes, newlineproteins and DNA oxidation resulting in hepatocyte injury. Herbal preparations have been newlineused for the treatment of liver diseases for a long time. Quality and standardization are newlinemaintained in these preparations. Eventhough herbal medicines are useful for the newlinemaintenance of health, the safety and efficacy of the herbal preparations are vital. The present newlinestudy has been undertaken to evaluate the toxicity profile and hepatoprotective activity of a newlinepolyherbal formulation. All the herbal ingredients were collected and checked for their newlineidentity as per AYUSH guidelines which found within the specified limits. newlineHydroalcoholic extracts of the individual plant materials were prepared, checked for newlinetheir active marker compounds using HPLC, combined in different ratio and it was taken as newlinethe final formulation. General physiochemical and safety parameters were analyzed for the newlinepolyherbal formulation by standard methods. Phytochemical marker compounds were newlinedetermined by HPLC/UHPLC method. The following phytoconstituents were found and they newlineare responsible for antioxidant, anti-inflammatory, antifibrotic and antihepatotoxic effects: newlinePicroside I - 6.29%, Picroside II - 7.45%, Picroside III - 1.13%, Cinnamic acid - 0.13%, newlineGallic acid - 0.43%, P-hydroxyacetophenone - 0.048%, Wedelolactone - 0.43%, newlineDemethylwedelolactone - 0.64%, Phyllanthin - 0.03%, Hypophyllanthin - 0.01%, Nirtetralin - newline0.04%, Swertiamarin - 0.16%, Mangiferin - 0.62%, Azadirachtin - 0.33% and Quercetin - newline0.67%. newlineThe polyherbal formulation (LVR05) was evaluated for its antioxidant activity using newlinedifferent in vitro methods which showed to have potent free radical scavenging activity.
