Abstract: Kidney stone formation also known as nephrolithiasis is the third most common newlinekidney disorder, affecting 12-15% of the global population. The present study was designed to investigate the protein make-up of human kidney stones and its intricate mechanism newlinein kidney stone formation. Decoding the etiology of lithogenesis at molecular level newlinecould aid in enhancing the therapeutic outcomes of this disease. To explore the human kidney stone organic matrix and its impact on the kidney stone pathology, surgically removed calcium oxalate kidney stones were collected from nephrolithiasis patients. It can be concluded that human kidney stone matrix proteins exert their antilithiatic potential through interacting directly with the COM crystals, consequently inhibiting the crystal-cell interaction, a pre-requisite to initiate inflammatory response. The present study provides molecular insights into the mechanism followed by urinary proteins to regulate the pathological mineralization in kidney. The findings from present study can help to fill in some of the gaps in the existing knowledge of the kidney stone formation, which could lead to better management of the disease. newline newline
