Abstract: | Acute and chronic gastro-intestinal infections and associated inflammatory newlinedisorders of various nature and magnitude are very common worldwide. The enteric newlinepathogens generate intestinal inflammation through breach of gut barrier system. newlineDisbiosis, a shift in peek populations of probiotics, commensals and pathogens has been newlinereported to be a leading cause of GI disorders. Restoration of the gut microbial peak populations through probiotics and herbal agents greatly influence metabolome which is central to the complete physiological and immunological response of an individual. Modulation of metabolome may therefore be an important tool that to prevent pathologies of gut immune system which no promising treatment is yet available. To evaluate the effect of cucurbits (Ls, Lc and Cp and probiotics (Lrh) on barrier reinforcement, inflammation and immuno-modulation in-vitro and in-vivo. Expression of gut barrier elements, Zonula Occludens (ZO-1) and mucin (MUC-2), inflammatory cytokines (NAP-3, COX-1, COX-2) and immunomodulatory cytokines IFN-and#947;, IL-17, IL-10, IL-4 and TGF-and#946;) were estimated through ELISA. Inflammatory molecule, Nitric oxide (NO) was estimated through Griess reaction. Adhesion of Lrh to epithelial cells and integrity of intestinal villi were studied newlinemicroscopically. Barrier strengthening effects of Ls, Lc and Cp may be attributed to various newlineantioxidants present in cucurbits like, polyphenols, carotenoids, and vitamins C. Antiinflammatory and immuno-modulatory activity of Ls, Cp and Lc may also be attributed newlineto their growth promoting effects on probiotics and microbial fermentation of dietary newlinefibers to short chain fatty acids (SCFAs) which are produced by probiotics through newlinemetabolism of dietary fiber. This study reveals the potential of cucurbits as non-toxic anti-inflammatory agents against gut ailments. The cucurbits, as non-toxic anti-inflammatory agents and nutrition enhancer can be helpful in inflamed systems. newline newline |
