In the present study, we implement a simple and high-throughput in vitro approch i.e. Bind-n-Seq to interrogate the DNA binding specificities of four seedspecific hexaploid wheat bZIP TFs and Arabidopsis TF Hy5. This information is predominantly read via and due to sequence-specific binding of transcription factors (TFs). The field of gene regulation has seen a tremendous leap in recent years in high-throughput methods and accuracy of protein DNA binding measurements. These techniques have been used in numerous studies such as producing better prediction of TF models, DNA-TF interaction landscape and improving the understanding of evolution of TFs. We expect these technologies and computational models to improve in coming years, advancing our ability to predict in vivo binding and to understand gene regulation in plants. newline
