Title: Isolation and Identification of Arsenic Transforming Bacteria and Study of Ground Water Quality in Mining Regions of Madhya Pradesh and Jharkhand India

Abstract: The arsenic (As) comprehensiveness in nature has aggravated the expansion newlineof arsenic fortification and detoxification components in microorganisms. Many newlinemicroorganisms discovered with ability to oxidize arsenite (As3+) into arsenate (As5+) newlineor reduce As5+ to As3+ newline. newlineIn this study, two bacterial strains designated as 3AB5 and 5AB5 were isolated from newlinethe soil samples collected from abandoned mining regions of Madhya Pradesh and newlineJharkhand, India. The arsenic concentration was determined in both water and soil newlinesamples. The physico-chemical properties of selected water samples were also newlinedetermined including water quality index. The quality of water samples studied were newlinefound moderate to very poor upon analysis for human use. Enrichment culturing newlinemethod was employed for isolating bacteria and further they were screened for their newlineredox ability. The isolated strains showed maximum growth at 300C, at pH 7.0 in newlinearsenic stressed minimal salt medium and were checked through UV-Vis newlinespectrophotometer at OD-620nm. Biochemical characterization of isolated strains was newlinecarried out with various biochemical tests. On phylogenetic analysis of selected newlinebacterial strains through MEGA-X, it was recorded that they have clear relationship newlineto the genus Bacillus. Further, they were also tested for transformation ability of newlinearsenic and gene identification was carried out in selected isolated strains using newlinepolymerase chain reaction. PCR analysis clearly indicates the presence of genes arsR, newlinearsB and arsC in the strain 5AB5 and gene related to oxidase activity aoxB was found newlinein 3AB5 strain, respectively. The result of this study clearly indicates that, even after newlineabandoning the mining activities concentration of arsenic increases in groundwater by newlinereducing ability of bacterial strains
