Abstract: newline Eleven indigenous potential probiotic bacteria obtained from traditional fermented newlinefoods of the Himachal Pradesh were screened for their functional and protective attributes. Eight newlineout of eleven indigenous probiotics were able to inhibit the activity of genotoxins (4-NQO and newlinefurazolidone) as observed by SOS chromotest. On the basis of in vitro antigenotoxic and newlineantimutagenic attributes, Lactobacillus plantarum (AdF10) and Lactobacillus rhamnosus GG newline(LGG) were selected to study their in vivo anticarcinogenic potential during 1, 2- newlinedimethylhydrazine (DMH) induced colon carcinogenesis. A significant reduction in total sialic newlineacid (TSA) and and#946;-glucuronidase activity was observed in DMH-treated animals supplemented newlinewith probiotics. Probiotics supplementation was also able to reduce the tumor incidences, tumor newlinemultiplicity and tumor size in DMH-treated groups. DMH treatment for a period of 16 weeks newlineresulted in a significant decrease in the activity levels of reduced glutathione (GSH), glutathione newlineperoxidase (GPx), glutathione-S- transferase (GST), superoxide dismutase (SOD) and catalase. newlineHowever, AdF10 (L. plantarum) and LGG (L. rhamnosus GG) supplementation to DMH-treated newlinerats, resulted in significant increase in the activity of these enzymes. Thus suggesting that newlineantioxidative potential of probiotics is one of the factors in providing protection against newlineexperimentally induced colon carcinogenesis. Probiotic supplementation to DMH-treated animals newlinemodulated protein expression of various genes involved in pathogenesis of colon cancer such as newlinep53, p21, COX-1, COX-2 and GLUT 2. The probiotics supplementation was also able to newlinemodulate immune system and behavior of animals. The viability studies on indigenous probiotic newlineisolates suggested that microencapsulation of probiotics in presence of different prebiotics could newlinebe a useful method for long term refrigerated storage
