dc.description.abstractAbiotic stresses in plants produce ROS, scavenged by anti-oxidant enzymes. Identification and characterization of SOD, CAT, APX, GPX and GR gene families in Brassica juncea, B. rapa and B. nigra was done using bioinformatics approaches, including chromosomal mapping, intron-exon organization, physico-chemical properties, sub-cellular location and 3D structure of proteins. Differentially expressed genes under abiotic-stresses in B. juncea and B. rapa were predicted using Trinityv2.03 and supported by the presence of cis-regulatory elements involved in abiotic stresses. Expression validation of candidate abiotic-stress responsive genes under drought and heat stresses was performed by qRT-PCR. Selected abioticstress responsive genes were cloned in expression vector pET-32a(+) and protein expression conditions were optimized. Enzyme and stability assays of purified recombinant BjuAFSD2a and BraAAPX2a proteins was done. newline
