Abstract: | The present research work was designed to screen Hypsizygus ulmarius and Pleurotus eous for newlinethe production of secondary metabolites, to study their therapeutic properties, and to elucidate newlinethe mechanism of action of their anti-obesity properties. The edible oyster mushrooms newlineHypsizygus ulmarius and Pleurotus eous were cultivated under in-house conditions on newlinevegetable waste and paddy straw. It was observed that maximum yield was obtained from newlineHypsizygus ulmarius as compared to Pleurotus eous (3539 g and 2171 g, respectively). Both newlinemushrooms proved to be rich sources of nutrients including moisture, ash, crude fat, protein, newlinecarbohydrates, and lipids. Moreover, Hypsizygus ulmarius was shown to contain higher newlinequantities of nutrients as compared to Pleurotus eous. The solvent extractions (acetone, newlineethanol, and water) of Hypsizygus ulmarius and Pleurotus eous showed high quantities of newlinesecondary metabolites such as alkaloids, polyphenols, flavonoids, tannins, melanin and newlinelovastatin which were evidenced from qualitative, quantitative, chromatographic and newlinespectroscopic studies. The acetone, ethanol, and water extracts of both mushrooms exhibited newline newlinetremendous anti-bacterial and anti-fungal activity. Their ethanolic extracts also expressed in- newlinevitro pancreatic lipase inhibitory activities at a concentration of 1 30 and#956;g/mL. Furthermore, in- newlinevivo studies in HFD (High Fat Diet)-induced obese mice (C57BL/6) revealed that newline newlinesupplementation of ethanol extracts of Hypsizygus ulmarius and Pleurotus eous in HFD- newlineinduced obese mice at dosage levels of 200 and 400 mg/kg body weight (C57BL/6) improved newline newlinetheir body weight, organ weight, serum biochemical parameters including serum triglycerides newlineand serum total cholesterol. In addition, the ethanol extracts of Hypsizygus ulmarius and newlinePleurotus eous improved mesenteric adipose morphometry and alleviated liver steatosis caused newlineby the accumulation of triglycerides within liver cells. Overall, the pharmacological potential, newlineanti-microbial, and anti-obesity properties of Hypsizygus ulmarius and Pleurotus eous could newlinebe attributed to the secondary metabolites present in them. The mechanism of action that newlineameliorates HFD-induced obesity in mice could be due to pancreatic lipase inhibitory activities newlineof the ethanolic extracts of Hypsizygus ulmarius and Pleurotus eous. newline |
