Abstract: Aim of the current work was to formulate and evaluate Chitosan-Catechol conjugated newlinepolymer followed by the design and development of Lafutidine loaded mucoadhesive pellets newlineand films. In this study conjugation and percentage of conjugation (DOC value) of Chitosan newlineand Catechol was inveterate by the NMR studies whereas other parameters like FTIR, DSC, newlineSEM and XRD were performed to test out the physiochemical properties of polymer. newlineInteraction of polymer with mucin was conceded through rheological studies which prove newlinethat polymer was strongly interacted with the mucus present in mucin layer. Mucoadhesion newlinepotential of Chitosan and Chitosan-Catechol conjugates was considered during In silico newlinemodeling using molecular mechanics stimulations and the obtained results were compared it newlinewith In vitro and ex vivo studies. Chitosan-Catechol conjugated polymer confirmed higher newlineenergy stabilization (and#8710;E and#8776; -65 kcal/mol) with mucin as compared it with pure chitosan with newlinemucin. Acute oral toxicity of the synthesized conjugates (Chitosan-Catechol) was performed newlineon swiss albino mice as per OECD guidelines. newline
