Characterization of Alkaline Pectin Lyase of Bacillus subtilis rsvr1 for Cotton Based Textile Industry Effluent Management

Abstract

Pectin is a hetero-polysaccharide, preset in the middle lamella of the primary newlinewall of the cotton. Pectin is a water-soluble molecule, but in plants it forms a complex newlinewith waxes, fats and proteins. This complex has water repellent property. In textile newlineindustry, it is necessary to remove this nonabsorbent layer before dyeing and newlinefinishing. Chemical processing of cotton generates a huge amount of pollutant and newlineharms the environment. newlineEnzymatic scouring can be an alternative to the above process. In this newlineinvestigation, we isolate, screened and produce pectin lyase form Bacillus subtilis newlinersvr1. Then, its applicability for scouring process was analyzed using Box and newlineBehnken design. COD and BOD values of chemical scouring as well as bioscouring newlinebath was evaluated. Maximum pectin lyase production was obtained with a medium newlinehaving composition of: 1.5 % of pectin, 0.2 % of ammonium chloride, 0.4 % of newlineK newline2 newlineHPO newline4 newline, 0.05 % MgCl newline at pH 9.0 and after 21 hour of incubation period. Purification newlineof enzyme was done in three steps: ammonium sulphate precipitation, ion exchange newlinechromatography and gel filtration chromatography. The enzyme was purified with a newline205.2-fold of purification and 291.4 U/g specific activity. The molecular weight of newlinepectin lyase was found to be 38 kDa. newline newline
