Abstract: ABSTRACT newlineBACKGROUND newlineIncreasing isolates of S. aureus with resistance to methicillin and decreased susceptibility to vancomycin has created concern for development of new anti-staphylococcal agents that kills resistant mutants. Plants have great potential as antimicrobial compounds having proven to have great therapeutic potential. newlineOBJECTIVE newlineScreening and molecular characterization of VISA and VRSA among the MRSA \isolates, and testing the efficacy of the purified compounds of Cannabis sativa (L.) and Allium sativum (L.) on these isolates. newlineMATERIAL AND METHODS newlineClinical samples were isolated from hospitalized patients in Rama Medical College Hospital and Research Centre Mandhan, Kanpur. Among the Staphylococcus aureus isolated the MRSA, VISA and VRSA were confirmed by MIC method and E-test. The detection of MecA and VanA gene was performed. newline newline newline newlineThe antibacterial activity ofplants was tested against these bacterial strains. newlineRESULTS newlineAmong the Staphylococcus aureus isolated from 140 samples, 85 strains belonged to MSSA, 55 strains were MRSA, of which 7 strains were VRSA in which 2 were confirmed to be VRSA by MIC method (along with E-test) and 8 strains were belonging with VISA category. newlineMolecular characterization and sequencing of MRSA isolates confirmed the presence of MecA gene. The MIC of pure phytomolecule of Cannabidiol (CBD) showed most effective 0.31 mg/ml as compared to pure Cannabinoid (mg/ml) of 0.40 mg/ml and crude Cannabis sativa extract of MIC 0.64 mg/ml whereas Allicin 0.42 mg/ml was more effective as compared to the Allium which showed MIC at 0.62mg/ml and crude extract of Allium sativum extract at 0.91 mg/ml in VISA and VRSA isolates. VanA gene was not detected among these isolates. newlineCONCLUSION newlineAbsence of VanA gene does not rule out that these strains are not VRSA or VISA. There is another hypothesis stating cell wall thickening is responsible for development of vancomycin resistance or may be other gene apart from VanA gene is responsible for the resistance , whereas plants can be used as potent age
